They also use bag seines to capture smaller prey species in marsh-edge and shoreline habitat—the seine is anchored to the boat and a biologist enters the water to open and close the seine.

They use otter trawls to sample finfish in larger inshore bays and waterways. During October through March, biologists use trammel nets to sample species such as red and black drum. Trammel nets are a type of gill net. A panel of smaller mesh is sandwiched between panels of larger mesh—this design allows for sampling of a large range of sizes and species.

In addition, biologists in some of the CSAs have been experimenting with electrofishing in the marsh and shoreline habitats to sample these same populations. Early results have shown that electrofishing may be a better sampling gear in certain locations.

Offshore sampling is often conducted as part of regional and national efforts, such as the Southeast Area Monitoring and Assessment Program SEAMAP , a cooperative state, federal, and university program to collect, manage, and disseminate fishery independent biological and environmental data from the marine waters of the southeastern United States.

LDWF biologists participate in a several SEAMAP projects, including sampling plankton , groundfish , and species caught with bottom longline and vertical line fishing gear.

Biologists collect plankton by towing fine mesh nets that taper into a collection tube next to a moving boat. Plankton is the collective name for microscopic eggs and larvae of fish and crustaceans that live adrift in the ocean currents. Neuston nets collect surface plankton, and bongo nets collect plankton found lower in the water column.

Biologists retrieve the nets after a set time, then sort and count the collected samples. Biologists tow a standard commercial shrimp trawl across muddy bottoms to sample an assortment of groundfish , other juvenile finfish, and invertebrates such as shrimp.

After each trawl, all of the specimens are collected, measured, weighed, and catalogued. By using a shrimp trawl, biologists are able to sample the juveniles of important species that cannot be caught in other surveys. Biologists set bottom longlines —long mainlines with baited droplines attached at even intervals—to sample larger animals along the bottom of the ocean.

After a set amount of time referred to as soak-time, they retrieve the lines and measure, weigh, and typically release each catch. The soak-time is short, increasing the likelihood that released fish will survive.

Some samples are kept for further study such as determining age and growth information. Various species of sharks make up the bulk of the catch, but red snapper, grouper, and other bottom dwellers occasionally show up.

In vertical line sampling , biologists use bandit reels—large spooled commercial fishing reels—to lower a line of hooks suspended above a large weight to the bottom around structures such as oil rigs. Hook sizes are varied to sample different sizes of fish. Bait type and soak-time are standardized, and biologists document all aspects of the catch for further analysis.

Red snapper is one of the most common catches in vertical line samples. In both fresh and saltwater habitats, biologists gather information about the current conditions at each sample site using a handheld meter with a digital probe that is lowered to various depths in the water column to read the water.

Based on sampling needs, these meters can read temperature , turbidity the amount of particulate matter in the water , dissolved oxygen , pH , and conductivity. By understanding the chemical and physical characteristics of the water, biologists can determine how certain conditions affect fish populations.

To date hundreds of thousands of core samples have been collected from floors of all the planet's oceans and many of its inland waters.

Coring began as a method of sampling surroundings of ore deposits and oil exploration. It soon expanded to oceans , lakes , ice , mud , soil and wood.

Cores on very old trees give information about their growth rings without destroying the tree. Cores indicate variations of climate , species and sedimentary composition during geologic history. The dynamic phenomena of the Earth's surface are for the most part cyclical in a number of ways, especially temperature and rainfall.

There are many ways to date a core. Once dated, it gives valuable information about changes of climate and terrain. For example, cores in the ocean floor, soil and ice have altered the view of the geologic history of the Pleistocene entirely.

Reverse circulation drilling is a method in which rock cuttings are continuously extracted through the hollow drill rod and can be sampled for analysis. The method may be faster and use less water than core drilling, but does not produce cores of relatively undisturbed material, so less information on the rock structure can be derived from analysis.

If compressed air is used for cutting extraction the sample remains uncontaminated, is available almost immediately, and the method has a low environmental impact. Contents move to sidebar hide.

Biologists use shoreline seines to sample black bass and sunfish species at night during the late summer. LDWF also samples freshwater mussel species around the state.

Mussels are an indicator species for the health of freshwater systems. Three-person crews of biologists sample mussels by wading in small streams and performing timed tactile searches for live mussels and shells. They also use SCUBA gear in waters deeper than one meter. In each of these areas, biologists conduct sampling focused locally on shrimp, crab, oysters, and coastal finfish like red drum and speckled trout.

Throughout the year, biologists use different sizes and types of trawls to sample shrimp in various habitats to determine season openings and closures. They SCUBA dive on designated sampling stations within each CSA. At each station, they randomly place a square meter frame on the oyster reef and collect all live and dead oysters, reef-associated organisms, and exposed reef material from the surface layer within the frame.

They replicate this process five times at each sampling station. Once the samples are back on the boat, biologists identify, separate, and count live and dead oysters, spat, fouling organisms, oyster predators, and hooked mussels; they also measure and sort the oysters.

Biologists use the data from square meter sampling in yearly oyster stock assessments. An oyster dredge is a metal frame with 10 teeth on one side that break up the ground as the dredge is pulled across the water bottom.

Dredged material is collected in a net or bag that is attached to the frame. Biologists examine the material for recent oyster spat larva settlement and signs of mortality.

LDWF samples saltwater finfish year-round along the coast. They also use bag seines to capture smaller prey species in marsh-edge and shoreline habitat—the seine is anchored to the boat and a biologist enters the water to open and close the seine. They use otter trawls to sample finfish in larger inshore bays and waterways.

Keep frozen samples solidly frozen at all times. Cool refrigerated samples, except shellfish and shell stock, in ice at °C and transport them in a sample chest with suitable refrigerant capable of maintaining the sample at °C until arrival at the laboratory. Do not freeze refrigerated products.

Unless otherwise specified, refrigerated samples should not be analyzed more than 36 h after collection. Special conditions apply to the collection and storage of shucked, unfrozen shellfish and shell stock 1.

Pack samples of shucked shellfish immediately in crushed ice no temperature specified until analyzed; keep shell stock above freezing but below 10C.

Examine refrigerated shellfish and shell stock within 6 h of collection but in no case more than 24 h after collection. Further details on sample handling and shipment may be found in the Investigations Operation Manual 5 and the Laboratory Procedures Manual 3.

The Investigations Operation Manual 5 contains sampling plans for various microorganisms. Some of those commonly used are presented here.

Because of the continuing occurrence of Salmonella in foods, sampling plans for these organisms have received the attention of committees of national and international organizations 6 , 7.

Each of these committees has recommended varying the number of samples from a particular lot of food according to the sampling category to which a food is assigned. Generally, the assignment to a sampling or food category depends on 1 the sensitivity of the consumer group e.

The selection of a sampling plan depends mainly on the first 2 criteria cited. The history of the food would be important in deciding whether to sample, i.

For the Salmonella sampling plan discussed here, 3 categories of foods are identified. Food Category I. Food Category II. Food Category III.

In certain instances, it may not be possible to fully conform to the sampling plan. Nonetheless it is still important to ascertain whether or not Salmonella is present in the suspect food. Therefore, the analyst should still try to analyze as many analytical units as is required for the food of interest, i.

Individual 25 g analytical units may be combined into g composites as described above unless otherwise indicated in Chapter 5, the OMA, or if the composited samples are not fully wettable.

Below are examples of situations that might confront the analyst. It does not apply to the collection of in-line process sample units at various stages of manufacture since those samples do not necessarily represent the entire lot of food under production.

The actual techniques involved in sampling are covered in the Investigations Operation Manual 5. A sample unit consists of a minimum of g and is usually a consumer-size container of product.

Take sample units at random to ensure that a sample is representative of the lot. When using sample containers, submit a control consisting of one empty sample container that has been exposed to the same conditions as those under which the sample was collected.

Collect more than one sample unit from large institutional or bulk containers when the number of sample units required exceeds the number of containers in the lot. A sample unit will consist of more than one container when containers are smaller than g e.

The numbers of sample units to be collected in each food category are as follows: Food Category I, 60 sample units; Food Category II, 30 sample units; Food Category III, 15 sample units.

Submit all samples collected to the laboratory for analysis. Advise the laboratory in advance of perishable sample shipments. Each sample should be mixed to ensure homogeneity before withdrawing a 25 g analytical unit.

The analytical units can be composited fifteen 25 g units into a g composite , unless otherwise indicated in Chapter 5 or in the OMA. Samples should be preenriched at a sample-to-broth ratio. For example, fifteen 1 lb bags of pasta have arrived for testing, but 5 of the bags are torn and unusable.

In this case, the analyst should only sample from the 10 intact bags. The contents of each intact bag should be mixed to ensure homogeneity before the analytical units are withdrawn. Since the analyst needs one g composite, ten For example, a single 10 lb wheel of cheese has arrived for testing.

Since cheese is a Category II food, thirty 25 g analytical units must be analyzed. These analytical units should be taken randomly from a wide variety of locations around the wheel.

If Salmonella is present in a food, then the odds of detecting it will be enhanced if two g composites are analyzed rather than a single 25 g analytical unit, as would be the case if the analyst were to treat the entire wheel as a single sample. For example, an 8 oz Almonds are a Category II food.

Category II foods require thirty 25 g analytical units g , so it is impossible to analyze the amount of almonds required by the sampling plan.

Fish Sampling Top 5 CSS Frrsh in Fresh sample program Ends. The same standard is used for any Frdsh and Reduced-cost grocery items sprays that contact the edible part of the Fesh. Riley, R. The method may be faster and use less water than core drilling, but does not produce cores of relatively undisturbed material, so less information on the rock structure can be derived from analysis.
